Abstract: The present invention relates to a microemulsion-based nano-delivery topical gel comprising a hydroalcoholic extract and/or a flavonoid-rich bioactive fraction of Duranta erecta Linn for enhanced wound healing. The invention provides a thermodynamically stable oil-in-water nano-microemulsion system incorporating Capmul MCM as the oil phase, Tween 80 as the surfactant, and Transcutol HP as the co-surfactant, which is optimized using Response Surface Methodology based on a Central Composite Design to obtain nanosized droplets with improved stability and performance. The optimized microemulsion containing approximately 10% (w/w) extract or fraction, 10% (w/w) oil phase, 30% (w/w) surfactant mixture, and 50% (w/w) aqueous phase is incorporated into a Carbopol 934 hydrogel with suitable excipients to form a smooth, homogeneous topical gel with acceptable pH, viscosity, and spreadability. The developed formulation exhibits enhanced solubility, dermal permeation, controlled release, and physicochemical stability over a three-month period, and demonstrates significant wound healing activity in an excision wound model in Wistar rats, including accelerated wound contraction, reduced epithelialization time, increased collagen deposition, improved antioxidant status, reduced lipid peroxidation, and enhanced tissue regeneration, thereby providing an effective herbal nano-delivery system for the management of acute and chronic skin injuries
