Abstract: The present invention relates to a bioactive polymeric herbal acne patch comprising Thespesia populnea (Portia fruit) extract incorporated into a pharmaceutically acceptable polymer matrix for the controlled topical delivery of bioactive phytoconstituents. The patch is fabricated by a solvent-casting technique using film-forming polymers comprising polyvinyl alcohol (PVA) and hydroxypropyl methylcellulose (HPMC) to obtain a flexible and adhesive dermal patch suitable for localized acne treatment. The developed formulation exhibited uniform physicochemical characteristics with a thickness of 0.28 ± 0.02 mm, weight of 123 ± 4 mg, folding endurance of 295 ± 12, and moisture content of 5.8 ± 0.3%, indicating good mechanical strength and stability. The formulation demonstrated sustained release of bioactive constituents for up to 10 hours, providing prolonged therapeutic action. The herbal patch exhibited significant antibacterial activity against Cutibacterium acnes with a zone of inhibition of 18 ± 1.2 mm, whereas the control patch showed no antibacterial activity. Cytocompatibility studies using L929 fibroblast cells confirmed excellent biocompatibility, with cell viability values of 95%, 90%, 85%, and 78% at concentrations of 25, 50, 100, and 200 µg/mL, respectively, remaining above the accepted safety threshold for topical formulations. The invention provides a natural, controlled-release topical drug delivery system exhibiting antimicrobial, antioxidant, and anti-inflammatory properties for effective management of acne vulgaris with improved patient compliance and reduced dependence on conventional synthetic therapies. The disclosed formulation is suitable for large-scale pharmaceutical and cosmeceutical manufacturing.
